TechniKrom®'s HPLC column is an industrial, portable dynamic
axial compression column for use at pressures up to 3000 PSI.
Dynamic axial piston compression allows for truly high performance
packing in minimal time. Placing constant pressure on the
piston gives it the dynamic capability of maintaining a consistent
homogeneous bed. The resulting elimination of voids and channeling
provides substantially increased chromatographic performance
and packed bed lifetimes. For lower pressure chromatographic
applications, use TechniKrom®'s LPLC column (0 –
150 PSI) or MPLC column (0 – 300 PSI), capable of packing
soft or hard gels at pressures as low as 3 – 4 PSI.

The design of the column assembly allows for quick packing
and unpacking. Portable columns with minimal footprint and
height also facilitate the performance of packing and unpacking
procedures in non-classified areas.
These columns are mounted on portable frames that allow 360°
rotation of the column to allow inversion during unpacking
if desired. To pack the column, the slurry inlet port is opened,
slurry is pumped into the column, the slurry inlet is closed,
and then a diaphragm pump is actuated to send the hydraulic
solvent (typically ethanol or water/ethanol, rather than machine
oil used in other vendors' columns) into the column to drive
the piston. This procedure is completed in a few minutes.
To unpack the column, the top flange is removed and the piston
is used to push the media out of the open column. The "cake"
can be directed to a tray or vessel by inverting the column.
These larger columns are mounted on legs (with wheels) with
the piston rod extending from the top. The packing media is
pumped into the column through the use of a slurry vessel
skid with an air diaphragm pump to quickly charge the media
through a column slurry inlet port. The piston is then pressurized
as in type 1 with a separate diaphragm pump using hydraulic
solvents. To unpack the column the lower flange is lowered
through the use of a portable jack and the piston is used
to push out the chromatography bed as a solid cake.

This unique distributor is an "active" design. Inlet
flow is directed through precisely machined horizontal channels
and vertically bored holes of precisely calculated diameters
to provide uniform distribution independent of frit or media
back pressure. It is incorporated into a low dead volume frit
assembly and provides minimal shear force, pressure drop and
band broadening. While TechniKrom® engineers each distributor
for the individual column ID, tests show that N or HETP vs.
flow rate is a straight line for all flow rates.
The leak detection quality assurance system involves a uniquely
designed port in the piston head between the main piston seals;
this system immediately signals any compromise of the seals.
If a seal is damaged, fluid will flow into this port and out
of the column through a separate tube and fitting on the upper
flange providing a visual alarm before any product is introduced
into the column. (It should be noted that the only way to
damage a seal is by improperly removing or replacing the piston;
a very rare occurrence.)
The piston has two main seals: one facing the stationary phase
and the other facing the hydraulic solvent. The seals incorporate
a u-cup design; they are composed of a FDA-approved high-grade
composition of PTFE with hardening agents added for mechanical
strength. To install the seal it is placed in hot water to
give flexibility and then simply pressed into the proper groove
by hand. The u-cup seal is an advanced design that expands
under liquid pressure, an improvement over the older chevron
seal designs where seal compression is achieved by turning
hard-to-reach bolts to manually expand the seals.
The end of the piston head (facing the stationary phase) is
outfitted with a scraper seal made of PTFE. The scraper seal
prevents packing material from approaching the main piston
seals. This is especially evident when unpacking the column.
The scraper seal a)prohibits any packing material from reaching
the piston seals, b) achieves an extremely clean column after
unpacking, and c) eliminates the dead volume between the column
wall and the upper piston head.

TechniKrom's frit design is that of "woven mesh,"
of welded multi-layered stainless steel screens. As a result,
our frit/distributor plate assembly has a lower dead volume
than the traditional sintered frit. The high surface area
and non-uniform pore structure of sintered frits results in
frequent clogging and higher back pressures.
The frame design is the most compact of any dynamic axial
compression column on the market today. Unlike other designs,
columns up to 80 cm ID can be wheeled through a standard doorway,
and columns up to 1 meter long can fit in a room with a ceiling
height of just 8 feet.
TechniKrom's columns incorporate a method of accessing the
lower piston seal, the piston scraper seal and the piston
frit without having to remove the top plate and piston from
the column. Thus, maintenance of these components can be achieved
in place, in a matter of minutes.
