TechniKrom designs and manufactures 316L stainless steel (and Hastelloy) LC columns ranging from 10cm to 2 meter internal diameter and up to 2 meter length. We design, build and test in accordance with ASME code calculations and FDA requirements and provide IQ/OQ validation documentation.

TechniKrom now offers a complete line of chromatography columns, dynamic axial compression or fixed. Our columns combine our leading edge flow distribution system with patent-pending hoist free design for ease of use. We offer bioprocess columns from 5 cm to 2 meter i.d. and HPLC and MPLC columns from 5 cm to 1.4 meter i.d.TechniKrom®'s process low-pressure (LPLC; 0 - 150 PSI) and medium-pressure (MPLC; 0 - 300 PSI) liquid chromatography columns use true dynamic axial compression. Axial piston compression allows for high performance packing in minimal time. Placing constant pressure on the piston gives it the dynamic capability of maintaining a consistent bed in light of packing media shrinkage, swelling, or channeling. This helps maintain an even long lasting bed that gives the highest possible resolution and sample distribution. Regulating this pressure controls the piston pressure; even the most fragile resins can be packed without crushing by setting the piston pressure as low as 3-4 PSI.

This unique distributor is an "active" design. Inlet flow is directed through precisely machined horizontal channels and vertically bored holes of precisely calculated diameters to provide uniform distribution independent of frit or media backpressure. It is incorporated into a low dead volume frit assembly and provides minimal shear force, pressure drop and band broadening.

Slurry Inlet and Outlet Ports
The LPLC and MPLC columns are equipped with ports along the side to aid in packing and unpacking. This design feature was created with two ideas in mind: (1) to decrease the time necessary to pack and unpack a column, and (2) to enable the user to pack and unpack a column within a "closed" system.

Leak Detection System
The leak detection quality assurance system involves a uniquely designed port in the piston head between the main piston seals; this system immediately signals any compromise of the seals. If a seal is damaged, fluid will flow into this port and out of the column through a separate tube and fitting on the upper flange providing a visual alarm before any product is introduced into the column. (It should be noted that the only way to damage a seal is by improperly removing or replacing the piston; a very rare occurrence.)

The two piston seals are composed of a FDA-approved UHMW-PE or virgin Teflon. The bottom seal faces the stationary phase, and the top seal faces air or hydraulic liquid (used to compress the bed dynamically). The coefficients of friction (µf) of these materials are very low and result in longer seal lifetime. This material is much better than the elastomers used in other column designs.

Front End Liquid/Scraper Seal
The end of the piston head (facing the stationary phase) includes a liquid/scraper seal made of FDA approved virgin Teflon or UHMW-PE. It performs as both a liquid seal and a wall scraper to leave the column completely clean during unpacking procedures, as evidenced by detailed visual inspections.

Woven Mesh Frit

Our frit design is that of "woven mesh," with welded multi-layered 316L stainless steel screens (polymeric or hastelloy versions are optional). As a result, our frit/distributor plate assembly has a significantly lower dead volume than the traditional sintered frit. The high surface area and non-uniform pore structure of sintered frits results in frequent clogging and higher back pressures.

Portability
The column design is the most compact of any dynamic axial compression column on the market today. Unlike other designs, columns up to 80 cm ID can be wheeled through a standard doorway, and columns up to 1 meter long can fit in a room with a ceiling height of just 8 feet.

Hoist-Free Maintenance
TechniKrom's LPLC/MPLC columns are designed for hoist free maintenance. Both of the column frits can be removed, cleaned, and replaced without the use of an overhead hoist or winch assembly.

• 316L stainless steel for all parts of the column including legs, frames and internal parts such as frits.
• Salt resistant passivated 316L stainless steel or Hastelloy available.
• All seals made of FDA approved elastomers; eg., PTFE, FEP, Viton.
• Sanitary design with minimized dead volumes and boroscopically examined orbital piping welds.
